The DNeasy PowerLyzer Microbial Kit is designed to isolate high quality genomic DNA from 1.8 ml of microbial culture. A variety of microorganisms including gram-negative and gram-positive bacteria, yeast, and bacterial and fungal spores have been tested successfully with this kit. The principal of this kit is to lyse the microorganisms by a combination of heat, detergent, and mechanical force using 0.1 mm glass beads. Use of our PowerLyzer 24 Homogenizer is recommended, but the kit can be used with any high powered bead beating instrument. The released DNA is then bound to a silica spin filter, washed and the DNA is recovered in certified DNA-free Tris buffer.

DNA was isolated from the gram positive bacteria  E. faecalis using the DNeasy PowerLyzer Microbial Kit (Glass Beads) and the DNeasy UltraClean Microbial Kit (Garnet Beads) using the PowerLyzer 24 Homogenizer at 2000 RPM for 5 minutes. 10 µl of each eluate was electrophoresed on a 1% agarose gel. A higher DNA yield was recovered using the glass beads in the PowerLyer™ UltraClean Microbial DNA Isolation Kit on a PowerLyzer 24 Homogenizer. The softer, garnet beads in the UltraClean Microbial DNA Isolation Kit are recommended for bead beating with a VortexGenie2.
